Overview
Psilocybe cubensis extract is a botanical drug substance derived from the *Psilocybe cubensis* mushroom, containing the psychoactive alkaloids psilocybin and psilocin. It is currently being investigated by the Centre Hospitalier Universitaire de Nantes (CHU de Nantes) in the Phase 2 LUCID-GAM clinical trial for the treatment of gambling disorder. The extract functions as a prodrug; once ingested, psilocybin is dephosphorylated into psilocin, which acts as a high-affinity partial agonist at the serotonin 5-HT2A receptor. This activation leads to downstream changes in cortical connectivity and neuroplasticity, which are thought to disrupt the maladaptive neural patterns associated with addictive behaviors and impulsivity, thereby facilitating a reset of the reward system and improving cognitive flexibility.
