PSMA+STEAP1 masked T cell engager is an investigational, tumor-activated, multi-specific T-cell engager (TCE) being developed by Xilio Therapeutics for the treatment of prostate cancer. Engineered using Xilio's proprietary Selective Effector-Enhanced Cell Engager (SEECR) platform, the molecule is designed to simultaneously target two highly prevalent prostate tumor-associated antigens: prostate-specific membrane antigen (PSMA) and six-transmembrane epithelial antigen of the prostate 1 (STEAP1). This dual-targeting approach aims to address tumor heterogeneity and minimize the risk of resistance due to antigen escape. The molecule features a masked CD3-targeting domain to prevent systemic T-cell activation and reduce systemic toxicities, such as cytokine release syndrome, alongside a built-in co-stimulatory domain (targeting CD28) to enhance the potency and durability of the T-cell response. Upon entering the protease-rich tumor microenvironment, the masks are cleaved, releasing the active, short half-life T-cell engager to selectively destroy prostate cancer cells.
