Overview

PSMA-617-S0456 is a dual-modality molecular probe designed for the targeted imaging of prostate cancer. It consists of the PSMA-617 ligand, which binds with high affinity to Prostate-Specific Membrane Antigen (PSMA), conjugated to the near-infrared fluorescence (NIRF) dye S0456 and the chelator DOTA. When labeled with Gallium-68 (68Ga), the probe enables preoperative PET/CT imaging to localize primary tumors and metastases. During surgery, the S0456 component provides real-time intraoperative fluorescence guidance, assisting surgeons in performing robot-assisted radical prostatectomy and identifying positive surgical margins or lymph node metastases. The agent is being evaluated in clinical trials at Xijing Hospital of the Airforce Medical University for its safety, feasibility, and diagnostic performance in patients with localized or locally advanced prostate cancer.
