Overview

This therapeutic vaccine is a combination immunotherapy developed by the H. Lee Moffitt Cancer Center and Research Institute for the treatment of prostate cancer, specifically in patients with biochemical recurrence (elevated PSA) following initial definitive treatment. The vaccine formulation consists of two peptide antigens: Prostate-Specific Membrane Antigen (PSMA) and T-cell receptor gamma alternate reading frame protein (TARP), which are tumor-associated proteins frequently expressed in prostate cancer cells. These antigens are combined with Poly IC-LC (Hiltonol), a synthetic double-stranded RNA that acts as a potent immunological adjuvant. Poly IC-LC stimulates the innate immune system by activating Toll-like receptor 3 (TLR3) and MDA5, which in turn enhances the maturation of dendritic cells and the induction of a robust T-cell mediated immune response against the targeted prostate cancer antigens. The vaccine is designed to train the immune system to recognize and eliminate residual or recurrent prostate cancer cells.
