Drug intelligence / Profile preview
PSMA anticalin radioligand is a preclinical-stage radiolabeled Anticalin protein being developed by XL-protein in collaboration with academic partners, including the Technical University of Munich (TUM). The agent consists of a human lipocalin 2 (Lcn2)-derived Anticalin engineered to bind prostate-specific membrane antigen (PSMA; also known as glutamate carboxypeptidase II) with high affinity. To optimize its pharmacokinetics for positron emission tomography (PET) imaging, the Anticalin is optionally modified using XL-protein's proprietary PASylation technology (conjugation to a proline/alanine/serine biopolymer) to extend its plasma half-life. The targeting protein is site-specifically conjugated to radiochelators (such as NODAGA, deferoxamine, or fusarinine C) and labeled with positron-emitting isotopes like Gallium-68 ($^{68}\text{Ga}$) or Zirconium-89 ($^{89}\text{Zr}$). This radioligand is designed for the non-invasive PET imaging of PSMA-positive tumors, particularly prostate cancer.
