Drug intelligence / Profile preview
PSMA-N064 is a preclinical theranostic, bimodal prostate-specific membrane antigen (PSMA) ligand. It consists of a glutamate-urea-lysine-based PSMA-targeting motif conjugated to the near-infrared (NIR) fluorophore/photosensitizer IRDye700DX and a DOTAGA chelator, which can be radiolabeled with indium-111 (111In). This dual-labeled agent is designed for intraoperative multimodal imaging (using SPECT/CT and near-infrared fluorescence) and targeted photodynamic therapy (tPDT) of prostate cancer. Upon binding to PSMA-expressing tumor cells and subsequent illumination with near-infrared light (690 nm), the IRDye700DX photosensitizer generates reactive oxygen species (ROS), inducing tumor cell apoptosis and DNA double-strand breaks. It has been evaluated in preclinical cell assays, mouse xenograft models, and ex vivo human prostate cancer tissue samples.
