Overview

**PSMA-pLipo** is a preclinical, prostate cancer-targeted liposomal gene-therapy candidate. It consists of liposomes carrying a prostate-specific antigen promoter-driven perforin expression vector and decorated with an anti-prostate-specific membrane antigen antibody for preferential delivery to PSMA-positive prostate cancer cells. Following delivery, PSA promoter activity is intended to drive perforin expression selectively in PSA-expressing tumor cells; perforin-mediated cytotoxicity is enhanced in the presence of cytotoxic immune cells. In reported in-vitro and 22Rv1DR xenograft studies, intravenous PSMA-pLipo inhibited growth of docetaxel-resistant, PSA-positive and PSMA-positive prostate cancer cells.
