Overview
**PSN-357** is a small molecule glycogen phosphorylase inhibitor developed by Prosidion (a subsidiary of OSI Pharmaceuticals) for type 2 diabetes and obesity. It prevents glycogen breakdown to glucose in the liver, rapidly lowering blood glucose levels, but its clinical development was discontinued due to poor selectivity between liver and muscle isoforms, leading to side effects like muscle fatigue. A Phase II trial evaluated its safety, tolerability, and exercise tolerance in diabetics.[1][5][7]
