PST-674 is a small molecule inhibitor of Dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A), currently under development by Pharmasum Therapeutics. DYRK1A is a kinase encoded on chromosome 21, and its overexpression is a primary driver of cognitive impairment and neurodegeneration in individuals with Down syndrome and Alzheimer's disease. By inhibiting DYRK1A, PST-674 aims to modulate the phosphorylation of tau and amyloid precursor protein (APP), potentially slowing the progression of dementia. The compound is designed to be brain-penetrant and is being advanced into clinical development for Down syndrome-related dementia, Alzheimer's disease, and Parkinson's disease dementia.
