PT-00114 is a synthetic peptide drug candidate developed by Protagenic Therapeutics for the treatment of stress-related neuropsychiatric disorders, including major depressive disorder, generalized anxiety disorder, post-traumatic stress disorder (PTSD), and opioid-related disorders. It is a 41-amino-acid analog of the naturally occurring brain peptide TCAP (Teneurin C-terminal associated peptide), designed to mimic its activity and counteract stress-induced hormonal effects. The mechanism of action involves inhibition of corticotropin-releasing hormone (CRH) signaling, thereby modulating the body's response to stress. Preclinical studies have shown that PT-00114 increases dendritic spine density in hippocampal regions and produces long-lasting changes in neuronal function by altering gene expression patterns. The drug has demonstrated excellent safety and tolerability in Phase 1 clinical trials with no clinically relevant adverse events reported up to doses of 1,000 micrograms administered subcutaneously. Its pharmacological profile includes rapid onset and long duration of action with weekly dosing expected[1][3][4][6][7].
