Overview

PT-401 is an experimental intranasal vaccine candidate designed to provide mucosal immunity against SARS-CoV-2, the virus responsible for COVID-19. Developed by researchers at the University of Connecticut (UConn), the vaccine utilizes a recombinant protein platform targeting the receptor-binding domain (RBD) of the viral spike protein. Unlike traditional intramuscular vaccines that primarily induce systemic IgG antibodies, PT-401 is specifically formulated for nasal administration to trigger a robust mucosal IgA response in the upper respiratory tract, potentially blocking viral entry and transmission more effectively. The vaccine is designed to be stable at room temperature, facilitating easier distribution in resource-limited settings. Preclinical studies have demonstrated its ability to induce high titers of neutralizing antibodies and provide protection against multiple variants of concern.
