Overview

PT1001B is a novel, selective small-molecule inhibitor of **type I protein arginine methyltransferases (PRMTs)**, with strong activity against PRMT1. It has been studied primarily for its role in **enhancing antitumor immunity** and overcoming resistance to anti-PD-L1 immunotherapies, particularly in cancers such as **pancreatic ductal adenocarcinoma** (PDAC). Mechanistically, PT1001B inhibits **PRMT1**, which leads to downregulation of PD-L1 expression on tumor cells, increases in tumor-infiltrating CD8+ cytotoxic T lymphocytes, and decreased PD-1+ leukocytes within the tumor microenvironment. These immune-modulating effects result in enhanced inhibition of tumor proliferation and increased induction of apoptosis, especially when combined with immune checkpoint inhibitors targeting PD-L1[1][3]. Preclinical models have shown PT1001B can reverse resistance to PD-L1 blockade and significantly inhibit tumor growth without observed toxicity in animal studies[1][3].
