Overview

PT403 is a long-acting, sustained-release formulation of the GLP-1 receptor agonist semaglutide, developed by Peptron for the treatment of type 2 diabetes and obesity. It utilizes Peptron's proprietary SmartDepot technology, which encapsulates semaglutide within biodegradable poly(lactic-co-glycolic acid) (PLGA) microspheres using an ultrasonic spray-drying process. This formulation is designed for once-monthly subcutaneous administration, aiming to reduce the injection burden compared to standard weekly semaglutide (e.g., Ozempic, Wegovy) and potentially improve patient adherence and tolerability. Preclinical data in obesity models and interim results from a Phase 1 pilot study in healthy subjects have demonstrated sustained efficacy and a favorable safety profile with fewer gastrointestinal side effects like nausea and vomiting compared to weekly semaglutide.
