Overview

PT404 is a long-acting, sustained-release formulation of tirzepatide, a dual glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptor agonist. Developed by Peptron using its proprietary SmartDepot™ ultrasonic spray drying technology, PT404 encapsulates tirzepatide within biodegradable poly(lactic-co-glycolic acid) (PLGA) microspheres. This formulation is designed to provide stable, sustained drug release over a monthly or bimonthly dosing interval, significantly reducing the injection frequency compared to weekly tirzepatide (Mounjaro/Zepbound). PT404 is currently in preclinical development for the treatment of type 2 diabetes and obesity, and is the subject of joint research and feasibility studies with Eli Lilly.
