PT502 is a first-in-class bispecific antibody designed to simultaneously target programmed death-ligand 1 (PD-L1) and CD47, two critical immune checkpoints exploited by cancer cells to evade immune surveillance. Developed by Phanes Therapeutics using its proprietary PACMAN platform, PT502 aims to overcome the limitations of existing CD47-targeted therapies, such as hematological toxicities like anemia and thrombocytopenia, by preferentially binding to CD47 on tumor cells that also express PD-L1. By blocking the PD-1/PD-L1 axis, PT502 restores T-cell activation, while its inhibition of the CD47/SIRPα "don't-eat-me" signal promotes macrophage-mediated phagocytosis of tumor cells. This dual mechanism of action potentially enhances anti-tumor efficacy through the synergistic activation of both innate and adaptive immune responses. PT502 is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors, including non-small cell lung cancer and gastrointestinal malignancies.
