Overview

PT586 is a bispecific antibody (bsAb) currently in IND-enabling development by Phanes Therapeutics for the treatment of glioblastoma (GBM). The molecule is engineered to simultaneously target a tumor-associated antigen (TAA) and CD47. CD47 is a cell surface protein that functions as a "don't eat me" signal by interacting with SIRPα on macrophages, thereby inhibiting phagocytosis. By dual-targeting, PT586 is intended to enhance the selective binding to tumor cells over healthy cells (such as erythrocytes), potentially improving the safety profile compared to monospecific CD47 inhibitors while promoting the immune-mediated destruction of glioblastoma cells.
