Overview

PTC-2105 (formerly known as SENO-201) is a first-in-class gerotherapeutic peptide being developed by Eos SENOLYTIX (following its reverse merger with Pulmatrix and acquisition of Senotherapeutix) for the treatment of sarcopenia and other aging-related diseases. As a senolytic agent, PTC-2105 is designed to selectively induce apoptosis in senescent cells, which are cells that have entered a state of permanent growth arrest but remain metabolically active and secrete pro-inflammatory factors. The peptide functions by disrupting the interaction between the transcription factor Forkhead box protein O4 (FOXO4) and the tumor suppressor p53. In senescent cells, FOXO4 sequesters p53 in the nucleus to prevent apoptosis; by interfering with this binding, PTC-2105 allows p53 to relocate to the mitochondria and trigger programmed cell death. This targeted clearance of senescent cells aims to reduce chronic inflammation and restore the regenerative capacity of skeletal muscle in elderly patients.
