PTC612 is an investigational, orally bioavailable small molecule inhibitor of the NLRP3 (NOD-like receptor protein 3) inflammasome, developed by PTC Therapeutics. The NLRP3 inflammasome is a multi-protein complex that triggers the release of pro-inflammatory cytokines IL-1β and IL-18 in response to various danger signals, and its dysregulation is implicated in a broad spectrum of inflammatory, metabolic, and cardiovascular diseases. PTC612 is designed to be highly potent and selective, featuring a chemical structure distinct from other NLRP3 inhibitors in development. As of mid-2026, it is undergoing Phase 1 clinical evaluation in healthy volunteers, including specific cohorts with obesity and cardiovascular disease to assess early pharmacodynamic activity.
