Overview

PTC844 is an investigational, orally bioavailable small molecule splicing modulator developed by PTC Therapeutics for the treatment of autoimmune disorders. It utilizes PTC's proprietary RNA-splicing platform to target the pre-mRNA of the CD40 ligand (CD40L, also known as CD154). By modulating the splicing process, PTC844 reduces the production of functional CD40L protein on the surface of activated T cells. This inhibition disrupts the CD40L-CD40 costimulatory interaction between T cells and B cells (or other antigen-presenting cells), which is a key driver of B cell activation, antibody production, and inflammatory cytokine release. PTC844 aims to provide a targeted approach to treating conditions like systemic lupus erythematosus (SLE) and other autoimmune diseases while potentially avoiding the complications associated with monoclonal antibodies targeting the same pathway.
