**PTD4-apoptin** is an experimental tumor-selective recombinant fusion-protein therapeutic comprising protein transduction domain 4 fused to apoptin, the VP3 protein of chicken anemia virus. PTD4 promotes cellular uptake, while apoptin preferentially accumulates in the nuclei of transformed cells, where it induces cell-cycle disruption and apoptotic cell death; in normal cells it is predominantly retained in the cytoplasm and is substantially less cytotoxic. The asset has shown antitumor activity in preclinical hepatocellular carcinoma, melanoma, cervical carcinoma, and gastric carcinoma models, including synergy with dacarbazine in melanoma models.
