PTEN lentiviral vector is a gene therapy construct designed to restore or overexpress the Phosphatase and tensin homolog (PTEN) protein in target cells. PTEN is a critical tumor suppressor that acts as a lipid phosphatase, dephosphorylating phosphatidylinositol (3,4,5)-trisphosphate (PIP3) to antagonize the PI3K/AKT/mTOR signaling pathway. Loss of PTEN expression is a hallmark of various malignancies, including hepatocellular carcinoma (HCC), prostate cancer, and glioblastoma, where it drives uncontrolled cell proliferation, survival, and metabolic reprogramming (the Warburg effect). By utilizing a lentiviral delivery system, the PTEN gene can be stably integrated into the host genome, providing long-term expression. While primarily utilized as a research tool to investigate tumor suppression and metabolic pathways, PTEN restoration via viral vectors is also explored as a potential therapeutic strategy in oncology and regenerative medicine.
