**PTEN mRNA@HA-LNP** is an investigational, localized mRNA replacement therapy comprising messenger RNA encoding phosphatase and tensin homolog delivered in a hyaluronate-conjugated lipid nanoparticle. The formulation uses hyaluronate-dimyristoyl glycerol to anchor hyaluronate on the nanoparticle surface, enabling skin penetration and preferential delivery to CD44-expressing melanoma cells following topical administration. Intracellular translation of the payload restores PTEN tumor-suppressor expression, suppresses PI3K/AKT signaling, promotes immunogenic cell death, and enhances antitumor immune activity. It has been reported in in vitro studies and melanoma-bearing mouse models, without identified clinical-trial development.
