PTG-007 + rituximab is an investigational combination therapy developed by PolTREG S.A. and its subsidiary Immuthera for the treatment of presymptomatic Type 1 Diabetes (Stage 1 and Stage 2). The combination consists of PTG-007, an autologous polyclonal T-regulatory (Treg) cell therapy, and rituximab, a chimeric monoclonal antibody targeting CD20. The therapeutic strategy involves using rituximab to deplete B cells, which are involved in antigen presentation and the production of autoantibodies, followed by the administration of PTG-007 to expand the pool of FoxP3+ regulatory T cells. This dual approach is designed to restore immune tolerance and protect pancreatic beta cells from autoimmune destruction, thereby delaying or preventing the onset of clinical symptoms in high-risk individuals.
