Drug intelligence / Profile preview
PTG-021 (anti-MOG CAR-Tregs) is an investigational autologous chimeric antigen receptor regulatory T-cell (CAR-Treg) therapy being developed by PolTREG and its subsidiary Immuthera for the treatment of neuroinflammatory and neurodegenerative diseases, including primary progressive multiple sclerosis (PPMS) and amyotrophic lateral sclerosis (ALS). The therapy consists of CD4+CD25+CD127- regulatory T cells genetically engineered to express a second-generation CAR targeting myelin oligodendrocyte glycoprotein (MOG), a key autoantigen in central nervous system demyelination. Administered intrathecally, PTG-021 is designed to migrate to sites of neuroinflammation, where MOG binding activates the CAR-Tregs to suppress autoreactive T-cell and antigen-presenting cell responses, thereby promoting immune tolerance and protecting myelin sheaths from autoimmune destruction. PTG-021 is prepared for Phase I/IIa clinical evaluation.
