PTG-100

PTG-100 is a first-in-class, orally stable, gut-restricted peptide antagonist that selectively targets the integrin alpha-4-beta-7 (α4β7). By blocking α4β7, PTG-100 inhibits the interaction with mucosal addressin cell adhesion molecule 1 (MAdCAM-1), thereby preventing memory T-cell trafficking to gut-associated lymphoid tissue. This mechanism is designed to reduce inflammation in diseases such as ulcerative colitis and celiac disease. Developed by Protagonist Therapeutics, PTG-100 was evaluated in clinical trials for moderate to severe active ulcerative colitis and celiac disease. Although initial phase 2b studies in ulcerative colitis were discontinued due to futility at interim analysis, subsequent blinded re-analysis suggested signals of efficacy and histologic remission at certain doses[2][3][5][6]. The drug has also been studied for its ability to prevent gluten-induced inflammatory injury in patients with celiac disease[4].
