Overview

PTG-630 is a first-in-class, orally available small molecule developed by Praetego as a neuroprotective agent. It acts as a potent inhibitor of advanced glycation end products (AGEs), targeting the oxidation of glycated proteins in the central nervous system. The drug’s mechanism involves capping redox metal ions, thereby preventing oxidative reactions and limiting synaptic breakdown and protein aggregation—key drivers of neurodegeneration. PTG-630 has demonstrated efficacy in preclinical models for protecting both the central and peripheral nervous systems, with functional benefits confirmed by biochemical markers such as amyloid, tau, and synaptic markers. Its primary indications under investigation include Alzheimer’s disease (AD), diabetic peripheral neuropathy (DPN), diabetic retinopathy, and chemotherapy-induced cognitive impairment ("chemo brain"). The compound shows good bioavailability, crosses the blood-brain barrier efficiently, and has a wide therapeutic window with minimal risk of drug-drug interactions[1][3][5][6][7][8].
