Drug intelligence / Profile preview
PTG-CARCIK-CD19 (also known as CMN-005) is an investigational, allogeneic, donor-derived chimeric antigen receptor (CAR) cytokine-induced killer (CIK) cell therapy targeting CD19. It is genetically engineered using the non-viral Sleeping Beauty transposon/transposase system (specifically the SB100X vector) to express a CD19-specific CAR containing CD28 and OX40 costimulatory domains and a CD3-zeta signaling domain. This non-viral manufacturing approach is designed to reduce the cost and logistical complexity associated with traditional viral vector-based CAR-T therapies. PTG-CARCIK-CD19 is being developed in collaboration by the University of Milano-Bicocca, Fondazione Tettamanti, and CoImmune (which merged with Formula Pharmaceuticals). It is currently in Phase I/II clinical trials for the treatment of B-cell malignancies, including B-cell precursor acute lymphoblastic leukemia (BCP-ALL), B-cell non-Hodgkin lymphoma (NHL), and chronic lymphocytic leukemia (CLL).
