Overview

PTGFRN CAR-T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy targeting Prostaglandin F2 Receptor Negative Regulator (PTGFRN), a transmembrane protein in the tetraspanin family. PTGFRN is overexpressed on glioblastoma (GBM) tumor cells but absent in nonmalignant human brain tissue, making it a potential tumor-specific antigen. Developed by researchers at Osaka University and Osaka National Hospital, the therapy utilizes a CAR construct derived from a monoclonal antibody library (specifically clone 5E17) screened against patient-derived tumor spheres. In preclinical studies, these CAR-T cells have demonstrated significant antitumor activity, including increased production of IL-2 and IFN-γ and reduced tumor growth in orthotopic xenograft models of glioblastoma.
