Overview

PTH(7-33)-CBD is a bone-targeted parathyroid hormone (PTH) antagonist designed to prevent breast cancer bone metastases. It consists of an N-terminally truncated analog of parathyroid hormone, PTH(7-33), fused to the inert collagen-binding domain (CBD) of ColG collagenase from *Clostridium histolyticum*. This fusion directs the delivery of the PTH antagonist to the bone matrix, where it blocks the action of parathyroid hormone-related peptide (PTHrP), a peptide secreted by breast cancer bone metastases that stimulates bone turnover and creates cavities for tumor growth. In preclinical models, PTH(7-33)-CBD has demonstrated the ability to reduce overall tumor burden in bone, decrease cortical bone destruction, and exhibit selective cytotoxicity against breast cancer cells without causing hypocalcemia or general toxicity.
