Overview

PTH-0301 is a monoclonal antibody targeting V-set domain-containing T-cell activation inhibitor 1 (VTCN1), also known as B7-H4. Developed by Protheragen, the therapeutic is designed to address solid tumors by blocking the B7-H4 immune checkpoint pathway. B7-H4 is frequently overexpressed on the surface of tumor cells and tumor-associated macrophages, where it serves to inhibit T-cell activation, proliferation, and cytokine production, thereby facilitating immune evasion. By binding specifically to VTCN1, PTH-0301 aims to alleviate this immunosuppression and restore the host's anti-tumor immune response. The program is currently in Phase I clinical development.
