Overview

PTH-0349 is a small molecule antagonist of the histamine H3 receptor (HH3R) currently in Phase II clinical development for the treatment of excessive daytime sleepiness (EDS). Developed by Protheragen, the compound targets HH3R, a G protein-coupled receptor primarily expressed in the central nervous system that acts as a presynaptic autoreceptor and heteroreceptor. By inhibiting HH3R, PTH-0349 promotes the release of histamine and other wake-promoting neurotransmitters, such as norepinephrine and acetylcholine, thereby enhancing alertness and wakefulness. It is being positioned as a potential therapy for sleep disorders where EDS is a primary symptom, such as narcolepsy and obstructive sleep apnea.
