Overview

PTH-0366 is a recombinant protein therapy designed to mimic the activity of human tissue kallikrein-1 (KLK1). Developed by Protheragen, the drug is intended for the treatment of acute ischemic stroke. KLK1 is a serine protease that cleaves low-molecular-weight kininogen to produce kinins, such as bradykinin, which interact with B1 and B2 receptors to induce vasodilation, inhibit inflammation, and provide neuroprotective effects. By restoring or enhancing KLK1 activity, PTH-0366 aims to improve cerebral blood flow and reduce tissue damage following an ischemic event. The program is currently in Phase I clinical development.
