A Pthlh inhibitor is a therapeutic agent designed to block the activity of Parathyroid Hormone-Like Hormone (PTHLH), also known as Parathyroid Hormone-Related Protein (PTHrP). In the context of pancreatic ductal adenocarcinoma (PDAC), PTHLH is identified as a key driver of epithelial-to-mesenchymal transition (EMT) and metastasis, particularly in tumors with p120 catenin loss. PTHLH functions as a secreted factor that binds to its cognate receptor, triggering cytosolic calcium (Ca2+) release and the subsequent activation of CaMKII signaling. Preclinical research indicates that genetic deletion or pharmacologic inhibition of PTHLH can impair tumor cell proliferation and migration, and reduce metastatic burden in vivo, suggesting that targeting the PTHLH-calcium signaling axis may offer therapeutic benefits for patients with aggressive pancreatic cancer.
