Drug intelligence / Profile preview
PTHrP neutralizing antibodies are a class of monoclonal antibodies designed to bind and neutralize parathyroid hormone-related protein (PTHrP). PTHrP is a tumor-derived factor that is frequently overexpressed in various solid tumors, including pancreatic, lung, breast, and head and neck cancers. It plays a critical role in driving tumor progression, osteolytic bone metastasis, cancer cachexia (including adipose tissue browning and muscle wasting), and humoral hypercalcemia of malignancy (HHM). By binding to the N-terminal region of PTHrP, these neutralizing antibodies block its interaction with the parathyroid hormone 1 receptor (PTH1R), thereby inhibiting downstream signaling cascades. Clinical-stage candidates in this class include BGM-2121, developed by BioGate Precision Medicine, which is currently being evaluated in Phase I clinical trials for advanced solid tumors.
