PTI-122 is a clinical-stage cytoprotective agent and pharmacokinetic modulator developed by Ratio Therapeutics. It is designed to be co-administered with PSMA-targeted radionuclide therapies, such as those based on Ratio's Trillium platform (e.g., [Ac-225]-Trillium-PSMA), to enhance the therapeutic index. The agent functions by modulating the biodistribution and pharmacokinetics of the radiopharmaceutical, specifically aiming to reduce off-target radiation exposure to healthy tissues like the salivary glands and kidneys, which are common sites of toxicity in PSMA-targeted treatments. By protecting these tissues, PTI-122 may allow for higher therapeutic doses of the radioligand to be delivered to metastatic prostate cancer lesions.
