PTK7 parental ADC is a preclinical antibody-drug conjugate (ADC) developed by Biocytogen. It consists of a fully human monoclonal antibody targeting protein tyrosine kinase 7 (PTK7) conjugated to the microtubule-disrupting cytotoxic agent monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline (vc) linker, typically with a drug-to-antibody ratio (DAR) of approximately 4. PTK7 is a tumor-associated antigen overexpressed in various solid tumors, including non-small cell lung cancer (NSCLC), breast cancer, and pancreatic cancer. PTK7 parental ADC is primarily utilized as a monovalent control and benchmark in preclinical studies to evaluate the synergistic efficacy and enhanced tumor selectivity of Biocytogen's bispecific ADCs, such as BCG017 (targeting PTK7 and EGFR) and BCG033 (targeting PTK7 and TROP2).
