PTK787 acts as an anti-angiogenic agent by inhibiting VEGFR-mediated signaling, aiming to reduce abnormal blood vessel growth and leakage. Verteporfin, when activated by light, generates reactive oxygen species to selectively destroy abnormal neovascular tissue. The rationale for their combination is to enhance anti-neovascular effects: verteporfin targets the destruction of existing pathological vessels, while PTK787 aims to suppress subsequent neovascularization driven by VEGF upregulation post-PDT[1][4].
