PTLS-001 (also known as PLS5) is a preclinical-stage small molecule mutual prodrug (Metual®) consisting of the chemotherapeutic agent paclitaxel covalently linked to the antidiabetic drug metformin. Developed by Primetime Life Sciences, PTLS-001 is designed to treat ovarian cancer by simultaneously delivering both active agents to the tumor site. Upon cleavage, paclitaxel acts as a microtubule stabilizer to inhibit cell division, while metformin targets mitochondrial complex I and PEN2 to activate AMPK, potentially overcoming chemoresistance and synergistically inhibiting tumor growth.
