PTM-101 is a proprietary flexible absorbable film device designed for the localized, sustained delivery of paclitaxel directly to the peritumoral area in patients with pancreatic cancer. The product enables continuous, high-dose administration of paclitaxel at the tumor site for up to six weeks, maximizing anti-tumor potency while minimizing systemic side effects typically associated with intravenous chemotherapy. Mechanistically, it acts as a local chemotherapeutic by releasing paclitaxel—a tubulin inhibitor—directly onto the tumor surface via laparoscopic placement. Preclinical and early clinical studies have shown that this approach achieves significantly higher drug concentrations in tumors compared to systemic dosing and leads to tumor reduction and improved survival in animal models. In first-in-human phase 1 trials involving patients with borderline resectable or locally advanced pancreatic ductal adenocarcinoma (PDAC), PTM-101 demonstrated favorable safety (no serious adverse events or systemic toxicity) and promising anti-tumor activity (tumor shrinkage observed in all treated patients). The chemotherapeutic agent remained localized within the pancreas without detectable systemic exposure[1][2][4][5][7].
