PTPRZ1-targeted CAR T-cell therapy is an experimental cellular immunotherapy designed to treat glioblastoma (GBM). It targets Protein Tyrosine Phosphatase Receptor Type Z1 (PTPRZ1), a cell surface antigen highly expressed on glioblastoma cells and glioblastoma stem cells, while being minimally expressed in normal adult brain tissue. One specific iteration, developed by researchers at the University of Geneva and Lausanne, utilizes an RNA-based CAR format (construct 471_28z) with a CD28 costimulatory domain to provide transient expression and potentially improve safety. Another program at Duke University employs an integrated single-cell discovery pipeline to optimize binders and CAR architecture for high sensitivity in low antigen-density environments. Preclinical studies have demonstrated potent antigen-specific cytotoxicity and bystander killing effects against PTPRZ1-negative tumor cells, leading to significant tumor growth delay in animal models.
