PTPRZ1-targeted CAR-T cells are an experimental cellular immunotherapy being developed for the treatment of glioblastoma (GBM). The therapy targets Protein Tyrosine Phosphatase Receptor Type Z1 (PTPRZ1), a surface antigen that is highly enriched in malignant glioma tissues, including glioma stem cells and various differentiation trajectories, while maintaining minimal expression in normal adult brain tissue. Developed through an integrated pipeline involving single-nucleus RNA sequencing and generative AI-driven antibody design, these CAR-T cells are engineered with TCR-mimetic architectures and modular costimulatory domains. This design aims to achieve high antigen sensitivity, improved functional persistence, and resistance to the immunosuppressive tumor microenvironment, addressing the challenges of intratumoral heterogeneity and cellular plasticity in GBM.
