Overview

PTRY is a trispecific antibody (tribody) developed by Chiome Bioscience that simultaneously targets the oncofetal tumor antigen 5T4, the T-cell receptor component CD3, and the immune checkpoint PD-L1. Part of the 53X tribody series, the molecule is engineered with a Fab domain targeting 5T4 and two single-chain variable fragments (scFv) targeting CD3 and PD-L1, respectively. Its mechanism of action involves functioning as a T-cell engager that facilitates the formation of an immune synapse between 5T4-expressing tumor cells and cytotoxic T cells, while concurrently blocking PD-L1-mediated immune suppression to enhance the anti-tumor response. Preclinical studies of the lead candidate, 53L10, have demonstrated potent anti-tumor activity and complete tumor regression in animal models. The drug is being developed for the treatment of solid tumors, including mesothelioma, lung cancer, and triple-negative breast cancer (TNBC).
