PTT-001 is an orally bioavailable small molecule pyroptosis inducer being developed by Pyrotech Therapeutics for the treatment of advanced solid tumors. It functions by activating Gasdermin E (GSDME), a protein that executes pyroptosis—a highly inflammatory form of programmed cell death. By triggering pyroptosis within the tumor microenvironment, PTT-001 promotes the release of pro-inflammatory cytokines and damage-associated molecular patterns (DAMPs), which enhances the recruitment and activation of tumor-infiltrating lymphocytes. This immuno-oncology approach is designed to convert immunologically "cold" tumors into "hot" tumors, potentially improving responses to checkpoint inhibitors and other therapies. PTT-001 is currently being evaluated in Phase 1 clinical trials.
