PTT-501 is a potent and highly selective small molecule antagonist of the Transient Receptor Potential Ankyrin 1 (TRPA1) channel, currently under development by Pyrotech Therapeutics (Beijing Yanming Biological Technology). TRPA1 is a non-selective cation channel primarily expressed in nociceptive sensory neurons, where it acts as a sensor for various noxious stimuli, including cold, mechanical stress, and reactive chemicals. In the context of chemotherapy-induced peripheral neuropathy (CIPN), TRPA1 is believed to be a critical mediator of the pain and hypersensitivity resulting from nerve damage caused by chemotherapeutic agents. By inhibiting TRPA1 activity, PTT-501 aims to provide a targeted therapeutic approach for managing the debilitating sensory symptoms associated with CIPN. The drug is currently being evaluated in Phase 1 clinical trials for safety, tolerability, and pharmacokinetics in healthy volunteers.
