PTT-936

PTT-936 is a first-in-class, orally bioavailable small molecule agonist of alpha-protein kinase 1 (ALPK1), developed by Pyrotech Therapeutics. It acts as an innate immune activator by targeting the cytosolic receptor ALPK1, which recognizes bacterial-derived ADP-heptose and triggers immune responses. By activating ALPK1, PTT-936 stimulates anti-tumor immunity and has demonstrated robust antineoplastic activity in preclinical studies both as monotherapy and in combination with immune checkpoint inhibitors. The drug is being developed primarily for the treatment of locally advanced or metastatic solid tumors, with additional potential applications in autoimmune diseases. Clinical trials are ongoing to evaluate its safety and efficacy.
