pTVG-AR is a DNA vaccine encoding the ligand-binding domain (LBD) of the human androgen receptor (AR). It is designed to stimulate an immune response against AR-expressing tumor cells, particularly in prostate cancer. The vaccine works by inducing antigen-specific CD8+ T cell responses targeting the AR LBD, which may delay disease progression and emergence of castration-resistant prostate cancer. Preclinical studies showed that pTVG-AR can augment anti-tumor immunity and prolong survival in animal models. Clinical trials have demonstrated that pTVG-AR is safe and immunologically active in patients with metastatic castration-sensitive prostate cancer (mCSPC), with evidence suggesting it may delay progression to castration resistance. The vaccine has also been studied in combination with androgen deprivation therapy (ADT) and immune checkpoint inhibitors such as nivolumab for high-risk or newly diagnosed prostate cancer[1][2][3][4][5][8].
