Overview

PTX-BD4-3 is a novel, small-molecule, non-peptide partial agonist of the tropomyosin receptor kinase B (TrkB) receptor, developed by PharmatrophiX. It is a derivative of the parent compound LM22A-4, optimized to exhibit improved blood-brain barrier permeability, a longer serum half-life, and enhanced oral bioavailability. PTX-BD4-3 mimics the neuroprotective effects of brain-derived neurotrophic factor (BDNF) by selectively binding to and activating TrkB, which triggers downstream survival signaling cascades such as the PI3K/Akt pathway. In preclinical studies, PTX-BD4-3 has demonstrated therapeutic potential for several neurological and neurodegenerative disorders, including Rett syndrome, Alzheimer's disease, stroke-induced thalamic injury, and post-traumatic epilepsy, by promoting synaptic resilience, reducing neuroinflammation, and preventing neuronal loss.
