PTX-COVID19-Variant is an investigational messenger RNA (mRNA) COVID-19 vaccine candidate being developed on Providence Therapeutics’ lipid nanoparticle-formulated mRNA platform as a variant-adapted version of its original PTX-COVID19-B vaccine. While detailed public data for PTX-COVID19-Variant are limited, it is expected to encode a modified full-length SARS-CoV-2 Spike protein incorporating amino acid changes found in variants of concern, in order to broaden or shift neutralizing antibody responses against circulating strains, while maintaining a similar mechanism to PTX-COVID19-B, which uses nucleoside-modified mRNA delivered by lipid nanoparticles to drive transient Spike expression, induce T helper 1–biased cellular immunity, and elicit high titers of neutralizing antibodies. The program is aimed at prophylactic immunization against COVID-19, leveraging prior preclinical and early clinical data from PTX-COVID19-B that showed robust humoral and cellular responses and protection in animal models, and favorable safety and immunogenicity profiles in humans.
