PU-PEI-miR145 is an experimental, nonviral microRNA delivery therapeutic consisting of tumor-suppressive miR-145 complexed with a cationic polyurethane-short branch polyethylenimine carrier. In glioblastoma CD133-positive cancer stem-like cells, delivered miR-145 directly suppresses the stemness-associated transcription factors OCT4 and SOX2, reducing tumorigenic and cancer stem-like properties, promoting differentiation, and increasing sensitivity to radiotherapy and temozolomide. Preclinical local intracranial administration suppressed orthotopic glioblastoma tumorigenesis and improved survival in immunocompromised mouse models.
