Overview

Pucilizumab (SG001) is a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. It functions as an immune checkpoint inhibitor by blocking the interaction between PD-1 on T cells and its ligands, PD-L1 and PD-L2, thereby restoring and enhancing T-cell mediated anti-tumor immune responses. Developed by Kelun-Biotech, pucilizumab is being investigated in various clinical trials, notably in combination with other targeted therapies such as the EGFR-targeted antibody vibepolimab. A significant area of clinical focus is its use in neoadjuvant settings for locally advanced, resectable EGFR-positive esophageal squamous cell carcinoma (ESCC), where it is evaluated alongside low-dose radiotherapy to improve pathological complete response and surgical outcomes.
